Unlike most other Marvel heroes, the modern Guardians of the Galaxy franchise revolves around being a melting-pot of Marvel universe properties. Because of this, characters here will be listed outside of their traditional rogues galleries if applicable. The Guardians are also criminal anti-heroes meaning some antagonists may overlap with allies and members.

- Like the MCU, the parks often mix up character franchises when finding villains to fight their respective heroes.
- Arnim Zola is a classic enemy of Captain America, used to fight Iron Man, Ant-Man and the Wasp in the parks.
- Taskmaster is an enemy of the Ant-Man, the Wasp and the Avengers who is used to fight Black Widow and Black Panther.
- Kang the Conqueror is an enemy of the Avengers and the Fantastic Four, affiliated with fighting Ant-Man and the Wasp in the films and the parks.
- The modern Guardians of the Galaxy franchise has the premise of being a melting-pot of different Marvel franchise characters, and in the parks uses enemies of the Avengers, Thor and the Eternals character Eson the Searcher.
- Several villains have their presences implied, though are not directly referenced:
- Of Black Panther's enemies:
- Erik Killmonger's presence is implied due to references to the events of the film Black Panther, namely its resolution where Kilmonger pressured them into opening up to the outside world.
- Of Captain America's rogues gallery:
- Advanced Idea Mechanics AKA AIM could be implied by references to Iron Man 3's plot.
- Of the Fantastic Four's enemies:
- Web Slingers a Spider-Man Adventure references experiments with the Negative Zone, the realm of the villainous Annihilus.
- Of the Iron Man rogues gallery:
- Obadiah Stane is alluded to by references to the plot of Iron Man (2008) and mention of his subordinate, William Ginter Riva.
- Aldrich Killian is implied by references to Iron Man 3's plot.
- The Mandarin can be inferred through the presence of the Ten Rings.
- Of Nick Fury and SHIELD/the Secret Warriors' enemies:
- The HYDRA leader Hive is indirectly referenced in Super Hero Headquarters where he the HYDRA facility which he was created at appears on a map.
- Zodiac's Zodiac Key makes a cameo in Mission: BREAKOUT!.
- Of Spider-Man's rogues gallery:
- Quentin Beck AKA Mysterio is referenced through post-cards in the queue for Web Slingers: A Spider-Man Adventure which allude to the plot of Spider-Man: Far From Home and in Avengers Assemble: Flight Force by mention of his subordinate, William Ginter Riva.
- Of Thor's rogues gallery:
- Hela is referenced through Jane Foster wielding Thor's shattered hammer.
- Surtur is alluded to through his Fire Dragon appearing in Monsters After Dark.
- The High-Evolutionary originates as a supporting-character from Thor comics who later became antagonistic towards characters like the Avengers. He is indirectly referenced in the parks as the creator of Rocket Raccoon who gave him the ID, "Subject 89P13", and may be indirectly referenced in Pym Test Kitchen through the meal-item, " Experiment 1517: Cardamom & Caramel Churro Cold Brew".
- Of the X-Men's enemies:
- The Crimson Bands of Cyttorak appear in Guardians of the Galaxy - Mission: BREAKOUT!. In the comics, Cyttorak is the source of power for the Juggernaut.
- Of Black Panther's enemies:
- One of several criticisms regarding Web Slingers: A Spider-Man Adventure has been its omission of having members of Spider-Man's classic rogues-gallery as the attraction's antagonists (such as how The Amazing Adventures of Spider-Man ride at Universal's Islands of Adventure features Doctor Octopus, Hobgoblin, Scream, Hydro-Man and Electro), instead having the Spider-Bots which are available for purchase in the gift-shop.
- In the comics, the Spider-Bots were originally created by Dr. Otto Octavius (Doctor Octopus) as opposed to Peter Parker.
